This document describes a method to determine the polysaccharide content of macroalgae biomass by monosaccharide determination after acid hydrolysis.
It specifies a method for the hydrolysis conditions to be used depending on the algae and a quantitative technique for determining the released monosaccharides and a calculation of polysaccharide content.
This method does not apply to the determination of polysaccharides from microalgae
Determining the polysaccharide content of macroalgae biomass is an important method in the industry for algae and algae products. A standardized protocol is needed for the hydrolysis conditions and measurement of the monosaccharide profiles for important polysaccharides like alginate, carrageenan and agar.
Therefore, this document is developed with the aim to give algae producers and algae products industries a recommendation for an analysis method for polysaccharides which can be used for macroalgae. The document will rely strongly on 2 key steps for the analysis. Optimized hydrolysis conditions (which need to be tailored according to the polysaccharide or algae of interest) and a quantitative technique for determining the released monosaccharides.
