ISO/NP 30024 (Ed 2) "Animal feeding stuffs - Determination of phytase activity"

Scope

This International Standard specifies the determination of phytase activity in feedingstuffs (including compound feeds, mineral complementary compound feeds, premixtures and feed additives) The method does not distinguish between phytase added as a feed additive and endogenous phytase already present in the feed materials.

The method cannot be used to evaluate or compare the in vivo efficacy of the phytase product. It is not a predictive method of the in vivo efficacy of phytases present on the market as they can develop different in vivo efficacy per unit of activity.

Actually, the method is suitable and validated exclusively for the determination of phytase activity and exclusively in complete feeds.

Purpose

1) adaptation of phytic acid (phytate substrate) specifications and editorial modifications Because referenced P0109 Sigma phytate substrate isn’t more available, we have to adapt the standard to future commercial products. For that, a description of specifications is necessary and verification procedures to check theses specifications will be useful.

Slight adaptations inside the whole text are necessary by consequences and especially the §10.3 about “Correction for phytic acid purity and water content”. In addition editorial modifications will be implemented if necessary

2) extension of the scope to premixtures and mineral feeds

New evidences from VDLUFA network indicate that the standard is applicable with small modifications to mineral feeds and premixture. Mineral feed and premixtures are ground, diluted with maize meal for matrix stimulation and extracted with acetate buffer. The solution is clarified by centrifugation or filtration. Afterwards the phytase activity is determined according to EN ISO 30024 from point 9.5 onwards on the level of a compound feed. 

It was fully validated during a collaborative study in 2011

Introduction of a § 9.6 paragraph in EN ISO 30024 describing dilutions of mineral feeds and premixtures.

Adding of an Annex B describing results of the 2011 VDLufa collaborative study on premixtures and mineral feeds (6 mineral feeds and 3 premixtures).

3) extension of the scope to feed additives

New evidences from VDLUFA network indicate that the standard is applicable to feed additives. Solid and/or liquid feed additives are extracted with acetate buffer. The solution is clarified by centrifugation or filtration. The cleared extraction solution is diluted with acetate buffer to a final activity between 0,06 – 0,16 U/ml. Afterwards the phytase activity is determined according to EN ISO 30024 from point 8.5 onwards on the same way as compound feed samples.

It was fully validated during a collaborative study in 2015

Adding of an Annex C describing results of the 2015 VDLufa collaborative study on feed additives (6 different types of products).

4) extension of the scope to samples containing a new 4a16 phytase feed additive In 2014-16, laboratories requested to the European Union Reference Laboratory for Feed Additives( EURL-FA) a study about applicability of ISO 30024 to feedingstuffs' samples containing a specific 6- phytase produced by Komagataella pastoris (registred as 4a16 in the European Union Register of Feed Additives). A specific intercomparaison study was performed in 2017-2018. That ring trial from EURL FA-NRL network indicates that the ISO method is fit for purpose to determine the phytase activity also in 3 complete compound feeds containing that 4a16 phytase and in 2 different formulation of the 4a16 feed additive.

It was fully validated during that collaborative study and recently published in November 2018.

details :

Adding, in the existing Annex A, results of the 2018 EURL-NRL collaborative study on 3 new compound feeds containing the specific 4a16 phytase. It will complete data already existing on compound feeds.

Adding in new Annex C, results of the 2018 EURL NRL collaborative study on feed additives (2 new different formulations of the 4a16 feed additive).
