This document specifies a method with the minimum technical requirements for DNA extraction of diatom samples, for use in DNA-based approaches dedicated to taxonomic identification of diatom communities, as well as ecological status and water quality assessments. The objective is to ensure high-quality DNA yields suitable for the subsequent molecular steps of DNA-based approaches (e.g. DNA metabarcoding). This document does not aim to cover the PCR, library preparation, sequencing, and data analysis steps of the workflow, which will be given in separate standards or technical reports.
Diatoms are a key component of benthic biofilms in aquatic ecosystems and are widely recognized as effective biological indicators for assessing ecological status and water quality [1,2]. In this context, DNA metabarcoding of diatom communities has been increasingly adopted as a complementary or alternative approach to traditional morphological identification. The successful application of metabarcoding, however, relies heavily on the quality and consistency of DNA extraction procedures. This document provides standardized requirements and guidelines for the extraction of DNA from diatom samples collected from biofilms, to ensure sufficient DNA yield and integrity for downstream molecular analyses. It supports harmonized practices across laboratories and contributes to improving the reliability and comparability of data produced through DNA-based assessments of diatom communities.
