Scope
This document specifies a laboratory method for the determination of the carotenoid content (with focus on fucoxanthin, lutein and beta carotene) in algae by extraction followed by HPLC-DAD (high pressure liquid chromatography coupled to diode array detector)
Purpose
Carotenoids are one of the major pigments present in algae and consist of the high value molecule classes carotenes and xantophylls. At this moment the commercially most interesting carotenoids from algae are fucoxanthin and lutein as xanthophylls and beta-carotene. Therefore the method will only be validated for these three carotenoids although it can be more generally applied to other carotenoids (except astaxanthin) when the right carotenoid standards are available.
The standard shall thus encompass the determination of the fucoxanthin, lutein and beta-carotene content in algae by means of extraction, HPLC separation and quantification by using the spectra obtained with a DAD (diode array) detector.
This determination shall enable that the carotenoids (with validation for fucoxanthin, lutein and beta-carotene) present at a significant level in the algal matrix are quantified in an accurate and reproducible way. The concentration of each carotenoid shall be expressed in mg/g dry weight. Moreover, the method described in the standard shall ensure a practical, quick, and safe technical approach, whose protocol details and all related know-how shall be easily and economically transferrable to all the sector stakeholders. The standard shall ensure this objective by a comprehensive and fully detailed description of all technical steps.
