CEN/TC 230 N 1262 , EN ISO 19040-2 Water quality - Determination of the estrogenic potential of water and waste water - Part 2: Yeast estrogen screen (A-YES, Arxula adeninivorans)

Scope

Highly sensitive and accurate determination of the estrogenic potential of aqueous samples for any salt content up to 3,5 %. The test method is applicable for the analysis of both influent and effluent waste water, for urban, industrial and hospital waste water. The method can be used for both filtered and unfiltered waste water without any sample pretreatment. It is applicable for any salt content up to 3,5 %. It can be used when it cannot be excluded that within the test series, survey or monitoring program the salt content of single samples may be above 0,5 %.

The test method provides both estrogen equivalent concentrations and DIL values, i.e. the dilution level from which on no more estrogen effect can be detected.

Without any extraction or enrichment of the water samples the detection of 1 ‐ 2 ng/l 17‐beta‐estradiol equivalents is possible, for fresh water, waste water and salt water.

Purpose

The test method allows the detection of estrogenic activity in the water cycle with very high sensitivity (1 - 2 ng/l 17-betaEstradiol). The test method can be used (1) for monitoring programmes for water quality, (2) for the surveillance of the performance of sewage treatment plants with regard to elimination of estrogenic substances, (3) as screening step before chemical analysis of single substances for any kind of water (e. g. as screening step for the surveillance of environmental quality standards), (4) for the analysis of aqueous extracts from sediments, particulate matter and soil and (5) for the analysis of eluates from enriched samples.

Due to the highly robust test organism the test method can be applied for a wide range of water matrices. The test organism Arxula adeninivorans is a highly robust and salt- and temperature-tolerant yeast. It tolerates temperatures up to 48 °C and very low pH-values up to 1,8.

To prevent any formations of antibiotic resistances in environment and to increase acceptance regarding legal requirements the test organism contains no antibiotic resistance markers.

As the Arxula adeninivorans cells are lyophilized the test can be conducted under highly standardized conditions in less than 26 h. No specific lab equipment for long-term cell cultivation is required. Lyophilized test organism is stored at 2 °C to 4 °C.

Precultivation (starter and main culture) is not needed for testing
