Scope

the qualitative detection of Atlantic salmon-specific DNA derived from foodstuffs and feedstuffs. It requires extraction of an adequate amount of PCR amplifiable DNA from the relevant foodstuffs and feedstuffs matrix and specific primer/probes unique for detection of Atlantic salmon material derived from Atlantic salmon (Salmo salar).

Purpose

Atlantic salmon (Salmo salar) and its related products hold a special place in the global market. They are highly sought after by consumers worldwide. Millions of tonnes of Atlantic salmon were harvested and farmed globally every year, with Norway, Scotland, and Chile serving as the main production regions. Additionally, North America, especially Canada and the East Coast of the United States, plays a significant role in Atlantic salmon production. Atlantic salmon is one of the most commercially successful fish species. Consumers highly value Atlantic salmon for its delicious taste and rich omega- 3 fatty acid content, making it one of the most popular health foods globally. Given the considerable demand and value associated with Atlantic salmon and its products, ensuring the authenticity of these products is crucial to meet market demand, safeguard consumer interests, and uphold the industry's reputation. This proposed technical specification describes a method to detect the Atlantic salmon-specific DNA sequence by targeting the specific conserved region of NADH dehydrogenase subunit 5 (ND5) gene (i. e. GenBank accession number LC012541.1) in the Atlantic salmon mitochondrion genome by real-time PCR. The method has been validated by 11 laboratories according to ISO 20813:2019. ISO/TS 20224:1~7: 2020, ISO/TS 20224:8~9: 2022 and ISO/TS 20224:10~11:2024, the detection methods of bovine, ovine, porcine, chicken, goat, horse, donkey, turkey, goose, duck and pigeon, have been published in the past years. The aim of the document is to add the Atlantic salmon DNA detection method to ISO 20224 as a new independent part.
